ChemDiv Platform Propels Neurodegenerative Drug Candidates

San Diego-based ChemDiv, Inc. has reached critical development milestones for its CNS-focused drug discovery platform, directly enabling clinical and preclinical progress for partners Brenig Therapeutics and Polku Therapeutics. The platform specializes in optimizing brain-penetrant small molecules to address complex neurological targets and chronic disease pathways.

Brenig Therapeutics is leveraging the platform to advance programs targeting the LRRK2 and NLRP3 pathways, both of which are central to neurodegenerative research. These candidates have now transitioned into clinical development, demonstrating the platform’s capacity to balance strict target selectivity with the rigorous pharmacokinetic requirements necessary for neurological indications.

Simultaneously, Polku Therapeutics reported success with its lead compound, POLK-38. In testing using the PS19 tauopathy mouse model—a standard for Alzheimer’s research—the compound demonstrated a clear impact on both spatial learning and brain tau pathology. POLK-38 functions by modulating prolyl oligopeptidase to restore protein homeostasis. Polku is set to share further research regarding its PREP-targeting platform at the Alzheimer’s Association International Conference on July 12, 2026.

Roman Timakhov, Research Director at ChemDiv, noted that integrating CNS exposure and selectivity into the initial design cycle allows partners to shorten the transition from early discovery to viable clinical candidates. This dual success highlights the firm's role in the pipeline of disease-modifying treatments for complex brain disorders.
